Abstract/Summary The extended peptide β-strand has been recognized as a common recognition motif for certain classes of biomolecular receptors. Chemically constraining molecules to adopt a β-strand conformation can substantially increase affinity for receptors that recognize β-strands or β-sheets. Many potential β-strand mimetics have not yet been recognized as such, and include a diverse array of rigid scaffolds and privileged structures, a few classes being surveyed in this update of the current literature on β-strand mimetics.
