Abstract/Summary Bacterial antigens of Coxiella burnetii were immunolabeled by freeze-drying, cryosubstitution and cryosectioning techniques. The three techniques were compared based on lipopolysaccharide profiles. Coxiella burnetii was prefixed in 3% glutaraldehyde, after which cells were pre-embedded in agarose on Durapore hydrophilic membrane prior to cryofixation. Cryosubstitution was carried out in a Reichert CS Auto Cryosubstitution apparatus. Samples dried in the Molecular Distillation Dryer were stabilized and transferred to a modified precooled cryobox. Polymerization of both cryosubstituted and freeze-dried samples were performed in L R Gold resin at 25 °C.
