Abstract A method is described for the purification of CEA from malignant ascitic fluid from a patient with serous cystadenocarcinoma of the ovary. Because CEA is relatively resistant to denaturation by cold and heat, the cryoprecipitable and heat-coagulable proteins in the fluid were removed before a 'mild' PCA extraction. Further steps of purification consisted of gel chromatography on Sephadex G150 and G200, Con A affinity chromatography and isoelectric focusing respectively. The final yield of CEA was 300 μg per 1. of fluid or 6.25 mg per kg total protein in the fluid. The CEA thus obtained appeared to possess similar physical and immunologic properties to colonic CEA.
