Abstract Alginates, ten other polysaccharides, and cell-wall preparations from Porphyra yezoensis are covalently dyed by Reactive Black 5. Dyed alginates resist cleavage by active preparations of guluronate lyase, indicating that dye-based assay cannot replace ultraviolet spectrophotometry in the guluronate lyase-linked assay for polymannuronan 5′-epimerase.
