Abstract/Summary The pressure of the helium blast and distance of the target tissue for microprojectile bombardment were studied using immature green cv. Davao Solo pawpaw fruits. In experiment 1, the effect of 4 different pressure levels (1000, 1500, 1800, and 2000 kPa) on transient gene expression was tested. In a second experiment, 3 distances (17.5 (the control), 15.0 and 12.5 cm) of the target somatic embryos from the filter containing the microprojectiles were tested. The zygotic embryos successfully produced somatic embryos after culture onto the somatic embryo induction medium. In experiment 1, the highest number of blue spots averaged 833, an indication of GUS expression, when the somatic embryos were bombarded at 1500-1800 kPa, while the target somatic embryos were ~17.5 cm away from the target. A lower pressure level, i.e. 1000 kPa gave low level of transient expression. Experiment 2 aimed to get high levels of GUS expression of ~1000 blue spots on the bombarded somatic embryos using a lower pressure. The blue spots on somatic embryos were highest at 1000 kPa when the target was 12.5 cm away from the filter unit. For stable transformation, a total of 160 bombardments using the antisense ACC synthase construct and controls were made. After one-month incubation of the bombarded somatic embryos on the pre-selection medium, their growth started to decrease. A further 2- to 3-months incubation on the full-selection medium caused death of many untransformed somatic embryos. Nine lines showed 2 bands while 5 lines showed 1 band, indicating hermaphrodite and female lines, respectively. None of the lines tested showed no band indicating that there are no male lines.
