目的 探讨肥胖抑制素(obestatin)对心肌细胞收缩、细胞内钙瞬变及细胞膜L-型钙通道的影响。
方法 采用单细胞收缩/荧光边缘探测系统及全细胞膜片钳技术观察obestatin对急性酶解分离大鼠心室肌细胞收缩及钙信号的影响。
结果 10-7mol/L obestatin增强大鼠心室肌细胞收缩幅度[(26.43±6.13)%]及细胞内钙瞬变幅度[(21.33±4.73%)],并增强ICa-L峰值。
结论 Obestatin开放心肌细胞膜上L-型钙通道,促进钙内流,增强大鼠心室肌细胞ICa-L及细胞内钙瞬变,进而增强大鼠心肌细胞收缩。

Objective To study the effects of obestatin on the contraction, calcium transient and L-type Ca2+ current (ICa-L) in rat ventricular myocytes.
Methods The extent of contraction and Ca2+ transient changes were recorded with the motion edge detection system, and L-type Ca2+ current was recorded with the whole cell patch-clamp technique.
Results Obestatin of 10-7mol/L increased the contraction amplitude of ventricular myocytes (26.43±6.13)% and Ca2+ transient amplitude (21.33±4.73)%, and enhanced ICa-L peak.
Conclusion Obestatin can strengthen contractility through increasing ICa-L and Ca2+ transient in rat ventricular myocytes.
