Abstract Lockednucleicacid (LNA) is one of the most prominent nucleicacid analogues reported so far. We herein for the first time report cleavage by restriction endonuclease of LNA–modifiedDNA oligonucleotides. The experiments revealed that RsaI is an efficient enzyme capable of recognizing and cleaving LNA–modifiedDNA oligonucleotides. Furthermore, introduction of LNA nucleotides protects against cleavage by the restrictionendonucleases PvuII, PstI, SacI, KpnI and EcoRI.
