The above paper reported the isolation of two furostanol steroidal saponins whose 25R absolute configuration was assigned on the basis of the geminal proton resonances ofH2-26 in methanol-d4, in accord with literature. However, subsequent acquisition of complete NMR data in pyridine-d5/D2O (~9:1) for 1 and 2 showed chemical shifts for H2-26 in 1H 3.51 and 4.10 ppm, Δδ0.59) and 2H 3.50 and 4.10 ppm, Δδ0.60) that were consistent with 25S (Δδ.0.57) rather than 25R (Δδ.0.48) stereochemistry. Additionally, the chemical shifts of H2-26 in 2 do not match those reported for protogracillin (25R) in pyridine-d5. Finally, the chemical shift of H3-27 (in pyridine-d5/D2O) was observed at δH 1.04 ppm (d, J¼6.7 Hz) for both 1 and 2, again more consistent with 25S (typically δH 1.01–1.05 ppm) as opposed to 25RH 0.98–1.03 ppm) configuration. The structures of the isolated saponins should therefore be revised to the 25S configured 1β-hydroxy protoneogracillin and protoneogracillin.
