Formatted abstract
A diversity of pharmacologic effects have been attributed to the industrial solvent, dimethyl sulfoxide (DMSO). Some of these effects are similar to the actions of non-steroidal anti-inflammatory drugs (NSAID), suggesting that DMSO may act by way of inhibition of prostaglandin synthesis. We tested the effects of DMSO on the oxidation of arachidonic acid by bull seminal vesicle (BSV) microsomes, a rich source of prostaglandin (PG) synthase, and on arachidonic acid-induced pulmonary platelet thrombosis in rabbits, a model for testing platelet anti-aggregation drugs. DMSO at concentrations of 8.6% and 36.7% (w/v) caused increased oxidation of arachidonic acid by BSV microsomes and at a dose of 1 g/kg did not protect against the induction of lethal pulmonary platelet thrombosis in rabbits by intravenously administered arachidonic acid (2 mg/kg). These observations are suggestive that rather than inhibiting PG synthase, DMSO may act as a reducing co-factor, and that DMSO does not inhibit thromboxane A2 synthesis by platelets.
