Abstract It appears that the genetic programming of multicellular organisms has been fundamentally misunderstood for the past 50 years, because of the assumption – largely correct in prokaryotes, but apparently not in the complex eukaryotes – that most genetic information, including regulatory information, is transacted by proteins. In humans and other complex organisms it appears increasingly likely that the majority of genetic information is embedded in the vast tracts of non-coding sequences that express regulatory RNAs to control the trajectories of differentiation and development, as well as the ontogeny and function of the brain. Thus what was dismissed as 'junk' because it was not understood may well hold the key to human complexity, variation and cognition.
